3.GAL4-UAS Transgene

Using attP-HiC31 transgenic technology, the gene of other species was transferred into the genome of Drosophila melanogaster. Through the GAL4-UAS binary regulatory system, the gene was expressed at high levels in Drosophila melanogaster and the phenotype can be analyzed. This method can simplify, mature, and standardize the study of gene function in non-model species.

Systemic and Temperature-sensitive Overexpression

Technical difficulties:When GAL4 is driven by a strong systemic promoter, exogenous genes downstream of UAS will be overexpressed in the whole body of Drosophila, which may result in toxicity, leading to decreased survival ability or even death of the flies.

Solution:Fungene has provided Tubp-GAL4,Tubp-GAL80ts strain. Transgenic flies can survive and expand at low temperatures, while also ensuring overexpression of exogenous genes after high temperature heat-shock.
